This new mutant, TMV [-25], was made using site-directed mutagenesis and resulted in an alteration of the coat protein translational start from AUG to AGA (Culver and Dawson, 1989b). This alteration left the other nucleotides of the coat protein open reading frame intact, including the original point mutation associated with the induction of HR, but prevented the production of the altered coat protein. Inoculation of mutant TMV [-25] onto expanded leaves of N. sy7vestris failed to induce the development of local lesions.
